Let CP's be Rs. 1000 each, their respective SP will be,1000 == 25%↑ ⇒ 1250 [person calculating profit on the CP]
1000 == 33.33%↑ ⇒ 1333.33 [The person calculating his profit on SP: 25% of SP = 33.33% of CP]
The difference turned out to be = 83.33. This has occured when we have assumed the CP as 1000. But, we are given difference of Rs. 100
So, on comparing,
83.33 = 1000
1 = $$\frac{{1000}}{{83.33}}$$
100 = $$\frac{{1000}}{{83.33}} \times 100$$ = Rs. 1200

He takes profit as 25% of CP
CP of SM1= x
SP of SM1 = y

CP of SM2 = z
SP of SM2 = y (because selling prices of two sales man are same)

5x/4 = 4z/3
x/z = 16/15

substitute x in (A)
16a + 25% (16a) = y
y = 16a + 4a ( here "4a" is profit for SM1)
y = 20a
substitute y, z in (B)
15a + 25% (20a) = 20a
15a + 5a = 20a ( here "5a" is profit for SM2)
Difference between profit of two sales mans is 100
So, 5a - 4a = 100
a = 100
Then,
y = 20a = 20 (100) = 2000

But it is given that the selling price is same in both the cases. why are you taking two different selling prices and same cost prices. In this case cost prices will be different.
Second person is calculating 25% of his profit on the Selling price.
And the selling price is
1000 + 25% of 1000 = 1250.
Now, 25% of 1250 = 312.5.
25% = 312.5
So, 100% = 1333.33
Thus, the actual profit on the Cost price would be,
(333.33*100)/1000 = 33.33%.
